Based on preclinical and Phase II clinical trials, ARA-290 (also known as Cibinetide) has shown potential benefits for neuropathy, chronic inflammation, tissue repair, and organ protection. Engineered to be a non-erythropoietic version of the hormone erythropoietin (EPO), ARA-290 activates the body’s innate repair receptor (IRR) without causing the risks associated with increased red blood cell production.
Being researched for the following potential benefits:
Neuropathic pain and nerve repair
-
- Reduces neuropathic symptoms: In clinical trials for diabetic and sarcoidosis-associated small fiber neuropathy, ARA-290 significantly improved neuropathic symptoms such as pain and tingling.
- Promotes nerve regeneration: Studies have shown that ARA-290 treatment can lead to an increase in small nerve fiber density, suggesting that it helps regenerate damaged nerve fibers.
- Inhibits pain pathways: It has been found to inhibit the TRPV1 channel, a pain receptor, which may provide direct analgesic effects in addition to its anti-inflammatory action.
- Reduces central inflammation: In animal studies, ARA-290 reduced spinal inflammation (microglia activation), which is linked to chronic neuropathic pain.
Chronic inflammation
- Modulates the immune system: ARA-290 helps downregulate pro-inflammatory cytokines and can calm an overactive immune response without causing broad immune suppression.
- Treats autoimmune conditions: Because of its immunomodulatory effects, it is being investigated for autoimmune diseases like sarcoidosis and rheumatoid arthritis.
Tissue protection and healing
- Accelerates cellular repair: By activating the innate repair receptor, ARA-290 triggers cellular responses that promote healing and protect against oxidative stress.
- Protects vital organs: Preclinical studies suggest protective effects on the kidneys, heart, and lungs from damage caused by ischemia or inflammation.
- Improves vascular health: It may enhance microcirculation and improve endothelial function, which benefits vascular health, especially in peripheral tissues.
Improved quality of life
- Reduces fatigue: In clinical trials for conditions like sarcoidosis, patients reported improvements in fatigue and overall physical functioning.
- Enhanced mobility: Studies showed increased exercise capacity in some patients with sarcoidosis-associated nerve loss.
Metabolic benefits: A Phase II study in type 2 diabetic patients showed that ARA-290 may improve hemoglobin A1c and lipid profiles.
